Description
(S)-M8-SPy is a new lanthanide chelating tag (M8) derived from an eight-fold, stereoselectively methyl-substituted DOTA. It is designed for paramagnetic labeling of biomolecules and can be covalently linked to the host molecule by a single disulfide bond. The steric overcrowding of the DOTA scaffold results in an extremely rigid, kinetically and chemically inert lanthanide chelator. Due to its exceptionally high stability and lanthanide affinity, M8 can be used under extreme chemical or physical conditions, such as those applied for protein denaturation or when it is undesirable for buffer or protein to react with excess lanthanide ions.
Uses
Used in Magnetic Resonance Imaging (MRI):
(S)-M8-SPy is used as a lanthanide chelating tag for caging lanthanides and other heavy metal ions in MRI. Its high stability and affinity for lanthanides make it suitable for use under extreme chemical or physical conditions.
Used in Structural Nuclear Magnetic Resonance (NMR) Applications:
(S)-M8-SPy is used as a lanthanide complex based on the DOTA cage in structural NMR applications. It has the ability to induce either paramagnetic relaxation enhancement or a pseudocontact shift (PCS) depending on the choice of the lanthanide, making it a valuable tool for studying molecular structures and interactions.
