LRRK2-IN-1, a benzodiazepine-based derivative, is a selective inhibitor of the Parkinson's disease kinase LRRK2. Leucine-rich repeat kinase 2 (LRRK2) is an enzyme that interacts with parkin, a ligase involved in the ubiquitin-proteasome system responsible for targeting proteins for degradation. Mutations in the LRRK2 gene, leading to increased kinase activity, have been strongly associated with the development of Parkinson's disease. LRRK2-IN-1 is a potent inhibitor of both wild-type and G2019S mutant LRRK2, with IC50s of 13 and 6 nM, respectively. It is selective for LRRK2 over a large panel of other kinases and promotes dephosphorylation, dissociation from 14-3-3 proteins, ubiquitination, and degradation of LRRK2. Additionally, LRRK2-IN-1 stimulates macroautophagy in H4 neuroglioma cells.

1234480-84-2 Usage

Uses

Used in Pharmaceutical Industry:
LRRK2-IN-1 is used as a therapeutic agent for the treatment of Parkinson's disease. Its application is based on its ability to selectively inhibit the LRRK2 kinase, which is implicated in the development and progression of the disease. By inhibiting LRRK2, LRRK2-IN-1 can potentially reduce the kinase activity associated with Parkinson's disease, leading to a decrease in dopaminergic cell death and an improvement in the symptoms and progression of the disease.
Used in Research Applications:
LRRK2-IN-1 is used as a research tool for studying the role of LRRK2 in Parkinson's disease and other related neurodegenerative disorders. Its high selectivity for LRRK2 over other kinases makes it a valuable compound for investigating the molecular mechanisms underlying the disease and for identifying potential therapeutic targets. Additionally, LRRK2-IN-1 can be used to assess the effects of LRRK2 inhibition on cellular processes such as macroautophagy, which may provide insights into the development of novel therapeutic strategies for Parkinson's disease and other neurodegenerative conditions.

Check Digit Verification of cas no

The CAS Registry Mumber 1234480-84-2 includes 10 digits separated into 3 groups by hyphens. The first part of the number, starting from the left, has 7 digits, 1,2,3,4,4,8 and 0 respectively; the second part has 2 digits, 8 and 4 respectively.
Calculate Digit Verification of CAS Registry Number 1234480-84:
(9*1)+(8*2)+(7*3)+(6*4)+(5*4)+(4*8)+(3*0)+(2*8)+(1*4)=142
142 % 10 = 2
So 1234480-84-2 is a valid CAS Registry Number.
