Three component condensation reaction as an efficient route for the synthesis of furo[2,3-d]pyrimidines

The condensation reactions of alkyl isocyanides with 1,3-dimethylbarbituric acid and aryl carboxaldehydes to afford 6-(alkylamino)-5-(3-aryl)-1,3- dimethylfuro[2,3-d]pyrimidine-2,4(1H,3H)-dione, in excellent yields under neutral conditions, are reported.

The one-pot three-component reactions of 1,3-dimethylbarbituric acid, aromatic aldehydes and alkyl or aryl isocyanides proceed smoothly at room temperature to give the corresponding 5-aryl-6-alkyl or arylamino-1,3- dimethylfuro [2,3-d] pyrimidine-2, 4 (1H,3H)-dione derivatives in good to excellent yields within 30 minutes in DMF. This three-component reaction represents a facile and efficient route to the described biologically interesting molecules.
