The invention relates to a preparation method of tert-butyl-7-bromo-5-oxa-2-azaspiro [3.4] octane-2-formate, and mainly solves the technical problem that no proper synthesis method is available for the compound at present. A three-step method is adopted for synthesis, and the method comprises comprises the following steps: 1, reacting 1-BOC-3-oxo-azetidine as a starting raw material with allyl bromide under the action of zinc powder to generate 3-allyl-3-hydroxy azetidine-1- tert-butyl carboxylate; 2, enabling the 3-allyl-3-hydroxyazetidine-1-tert-butyl carboxylate to react with liquid bromineso as to generate tert-butyl-3-(2, 3-dibromopropyl)-3-hydroxyazetidine-1-formate; and 3, carrying out a reaction on the tert-butyl-3-(2, 3-dibromopropyl)-3-hydroxyazetidine-1-formate under the condition that potassium carbonate is adopted as an alkali to generate the tert-butyl-7-bromo-5-oxa-2-azaspiro [3.4] octane-2-formate. The tert-butyl-7-bromo-5-oxa-2-azaspiro [3.4] octane-2-formate is a useful intermediate or product for the synthesis of many medicaments.
