Synthesis of 1-(3-substituted-2-hydroxypropyloximino)benzocycloalkanes as potential antiamoebic agents

Epoxypropylation of benzocycloalkan-1-oximes (1-3 and 5) with epichlorohydrin gives the corresponding 1-(2,3-epoxypropyloximino)benzocycloalkanes (6-9).Condensation of 6-8 with NaN3 and of 6-9 with various amines affords 1-(3-azido/substituted amino-2-hydroxypropyloximino)benzocycloalkanes (13-30).Acetylation of 13 and 20 gives their acetoxy derivatives (31 and 32).Alkylation of 2 with 1,3-chlorobromopropane yields the 3-chloropropyloximino derivative (33) which on reaction with various amines furnishes 1-(3-substituted aminopropyloximino)tetralins (34-36).Compound30 shows in-vitro antiamoebic activity at 31.25 μMg/ml concentration against E. histolytica.None of the 11 tested compounds shows in-vivo antiamoebic activity.
