Aerobic glycolysis, originally identified by Warburg as a hallmark of cancer, has recently been implicated in immune cell activation and growth. Glucose, the starting material for glycolysis, is transported through the cellular membrane by a family of glucose transporters (GLUTs). Therefore, targeting glucose transporters to regulate aerobic glycolysis is an attractive approach to identify potential therapeutic agents for cancers and autoimmune diseases. Herein, we describe the discovery and optimization of a class of potent, orally bioavailable inhibitors of glucose transporters, targeting both GLUT1 and GLUT3.
TREATMENT OF INFLAMMATORY CONDITIONS AND AUTOIMMUNE DISEASES WITH GLUCOSE UPTAKE INHIBITORS
-
Paragraph 0143; 0144, (2018/11/22)
This invention provides methods of treating inflammatory conditions and autoimmune diseases in a mammal, comprising administering to the mammalian subject in need thereof a therapeutically effective amount of a compound, or pro-drug thereof, or pharmaceutically acceptable salt or ester of said compound or pro-drug, wherein the compound is an inhibitor of glucose uptake. In particular aspects, the present invention provides a method of treating inflammatory conditions and autoimmune diseases in a mammal, and preferably a human, in which the glucose uptake inhibitor modulates the glucose transport of GLUTl and GLUT3.
TREATMENT OF GVHD
-
Paragraph 0223, (2015/11/18)
The invention relates to treatment of graft versus host disease (GVHD) using compounds that inhibit ROCK2. In preferred aspects, the present invention provides methods for the treatment of GVHD, including chronic GVHD ( cGVHD) using compounds having the formulae l-XXV, as set forth herein.
TREATMENT OF OCULAR DISORDERS
-
Paragraph 0195, (2014/04/18)
The invention provides methods of treatment of ocular disorders, including ocular disease with an angiogenic component. In certain embodiments, the treatment comprises administration of a ROCK2 inhibitor and an angiogenesis inhibitor. In certain embodiments, the ROCK2 inhibitor is ROCK2 selective. In certain embodiments, the angiogenesis inhibitor is a VEGF antagonist, for example, and VEGFR2 antibody.
