Copper complex of polyglycerol anchored to graphene oxide as a recyclable nanocatalyst for sonochemical green synthesis of naphthoquinones

In the present study, Cu(II) immobilized on hyperbranched polyglycerol functionalized graphene oxide catalyst was prepared as a novel, green, highly efficient, and reusable heterogeneous catalyst. Then, the synthesis of naphthoquinone derivatives was performed using Go-HPG-IA-Cu(II) as a catalyst under ultrasound irradiation to afford the desired products in high yields and short reaction times. This catalyst was easily recovered and reused several times without significant loss of catalytic activity.

A new Bi(iii) immobilized triazine dendrimer-stabilized magnetic nanoparticle (Fe3O4@TDSN-Bi(iii)) catalyst was prepared and characterized by FT-IR, TGA, elemental analysis, FE-SEM, TEM, VSM, XRD, XPS and lCP-OES techniques. This catalyst was used efficiently for the preparation of different aminonaphthoquinones via a one-pot three-component reaction of lawsone, aldehydes and amines in EtOH as a green solvent at room temperature. This catalytic system also showed excellent activity in the synthesis of symmetric and unsymmetric bis-aminonaphthoquinones from dialdehyde and/or diamine in high yields and purity via an easy work-up procedure. The easy recovery and reusability of the catalyst make this method attractive for green chemistry.

A new Cu(II) immobilized on hyperbranched polyglycerol (HPG) functionalized graphene oxide catalyst was prepared and characterized by FT-IR, TGA, FE-SEM, XRD, and ICP-OES techniques. This catalyst was used efficiently for the preparation of aminonaphthoquinones via one-pot three-component condensation reaction under solvent-free conditions. The catalyst could be easily recovered and reusable several times without a significant loss of activity, which make this method attractive and in a close agreement with green chemistry.

An efficient one-pot protocol for the synthesis of 2-((substituted amino)(4-phenyl)methyl)-3-hydroxy-naphthalene-1,4-dione derivatives has been developed by the three-component reaction of 2-hydroxy-naphthalene-1,4-dione, aromatic aldehydes and anilines/heterocyclic amines using montmorillonite K-10 as a catalyst. The advantages of this method include short reaction time, excellent yield and easy work-up.

Hydroxynaphthoquinones such as lawsone (2-hydroxy-1,4-naphthoquinone) have proven to be effective antifungal agents. These compounds were tested for antifungal activity against yeast standard and clinical strains by the broth microdilution method. Among the synthetic lawsone derivatives, 2-hydroxy-3-((2-hydroxyphenyl)(pyrrolidin-1-yl)methyl)naphthalene-1,4-dione, 2-hydroxy-3-(((4-nitrophenyl)amino)(phenyl)methyl)naphthalene-1,4-dione and 2-hydroxy-3-((2-hydroxyphenyl)((4-nitrophenyl)amino)methyl)naphthalene-1,4-dione showed high activity against Candida albicans ATCC 10231, with minimal inhibitory concentrations (MICs) and minimal fungicidal concentrations (MFCs) ranging from 20 to 330 and from 80 to 330 μg mL-1, respectively. Moreover, they also showed a mechanism of action on exogenous ergosterol. Therapeutic concentrations (CC50) of 2-hydroxy-3-((2-hydroxyphenyl)(pyrrolidin-1-yl)methyl) naphthalene-1,4-dione, 2-hydroxy-3-(((4-nitrophenyl)amino)(phenyl)methyl)naphthalene-1,4-dione and 2-hydroxy-3-((2-hydroxyphenyl)((4-nitrophenyl)amino)methyl)naphthalene-1,4-dione were 52.81, 52.58 and 85.94 μg mL-1, respectively, which can be considered moderate or low. In addition, docking studies showed that these compounds had similar binding energy to standard ketoconazole, which are recognized as the molecular target by van der Waals interactions. Furthermore, they are under Lipinski's rule of 5 with a druglikeness score better than ketoconazole and nystatin. These findings suggest that 2-hydroxy-3-((2-hydroxyphenyl)(pyrrolidin-1-yl)methyl) naphthalene-1,4-dione, 2-hydroxy-3-(((4-nitrophenyl)amino)(phenyl)methyl)naphthalene-1,4-dione and 2-hydroxy-3-((2-hydroxyphenyl)((4-nitrophenyl)amino)methyl)naphthalene-1,4-dione have potential as leading compounds against human fungal infections.

A clean, efficient and facile one-pot protocol was developed for the synthesis of a series of new aminonaphthoquinones derived from 2-hydroxy-1,4-naphthoquinone (lawsone) by three-component Mannich reaction using catalytic amount of p-TsOH in CH3CN, at room temperature. At the present work, we improved the yield and significantly reduced the reaction time for several Mannich reactions with different amine and aromatic aldehydes using a non-expensive, mild catalyst and suitable solvent.
