The invention relates to a method for preparing a chiral spiro isotetrortic acid derivative. The chiral spiro isotetrortic acid derivative which is almost optically pure is obtained by performing multi-step tandem reaction on low-cost and readily-available raw materials isatin derivatives and alpha-keto ester under the condition that a cinchonic alkali derivative is taken as a catalyst and dichloromethane is taken as a solvent. The reaction raw materials are low in cost and readily available, convenience in catalytic reaction operation is ensured, and only the raw materials and the catalyst are required to be dissolved in the optimized organic solvent at low temperature without stirring and TLC (Thin Layer Chromatography) reaction monitoring.
