The invention provides a synthesis method of 1, 5-dimethylpyrimidine-2 (1H)-ketone, and relates to the field of organic synthesis. According to the synthesis method, 2-chloro-5-methylpyrimidine and anacid are stirred in an oil bath to react to obtain a 5-methylpyrimidine-2 (1H)-ketone acid salt, and the 5-methylpyrimidine-2 (1H)-ketone acid salt reacts with an alkali and methyl iodide in an organic solvent to prepare 1, 5-dimethylpyrimidine-2 (1H)-ketone. The synthesis method provided by the invention has the advantages of easily available raw materials, mild reaction conditions, high selectivity, good operability of post-treatment and high product purity, and is suitable for large-scale industrial production.
