The pyrimidinethione 1 has been reacted with hydrazine hydrate and acid hydrazides gave compounds 2 and 3. And with primary amines yielded compound 4. Also, the behavior of compound 1 towards carbon electrophiles, e.g., phenylisocyanate, acrylonitrile, acetic anhydride, dimethyl sulphatee and ethyl chloroacetate has been investigated and yielded compounds 5a-e, respectively, the ester 5e reacted with hydrazine hydrate and primary amines and gave the hydrazide 5f and compounds 6a-d. The behavior of the hydrazide 5f towards nitrous acid and carbonyl compounds has been described and gave the azide 5g and compounds 7 and 8. The reaction of compound 1 with secondary amines under Mannich reaction condition and oxidation of 1 with HNO2/CH3COOH has been discussed.

Reaction of 4-chlorobenzylidene-2,4-dimetyl acetophenone and 4-chlorobenzylidene-2-methoxy-4-methyl acetophenone with thiourea gave the corresponding pyrimidin-2-thione derivatives. These pyrimidin-2-thione derivatives reacted with a variety of nucleophiles and electrophiles to give substituted pyrimidiues, mercaptopyrimidines, furanones, and pyridazines.
