Alemtuzumab, also known as Campath, is a humanized monoclonal antibody (Campath-1H) that targets the 21to 28-kDa cell surface glycoprotein CD52. CD52 is expressed on the surface of normal and malignant B and T lymphocytes, NK cells, monocytes, macrophages, and tissues of the male reproductive system. Alemtuzumab is an IgG1 κ form with humanized variable and constant regions and CDRs from a rat monoclonal antibody, Campath-1G. It was first introduced in the US for the treatment of B-cell chronic lymphocytic leukemia (CLL) in patients who have been treated with alkylating agents and have failed fludarabine therapy.

Used in Antineoplastic Applications:
Alemtuzumab is used as an antineoplastic monoclonal antibody for the treatment of B-cell chronic lymphocytic leukemia (CLL) in patients who have been treated with alkylating agents and have failed fludarabine therapy. It binds to CD52, a nonmodulating antigen present on the surface of essentially all B and T lymphocytes, most monocytes, macrophages, and NK cells, and a subpopulation of granulocytes. The proposed mechanism of action is antibody-dependent lysis of leukemic cells following cell surface binding.

In a phase III clinical trial with 93 fludarabine-resistant CLL patients, treatment with alemtuzumab gave a positive response in 31 patients with 2 complete remissions and 29 partial remissions. This study demonstrated a higher response rate in patients having malignancy confined to the blood and bone marrow.
