Synthesis of certain alkyl and aralkyl-4-[(alkylamino)methyl]-2-(4- methoxybenzyl)pyrrolidin-3-ols of anticonvulsant and neuroleptic profile

The synthesis of a 1-alkyl and aralkyl-4-[(alkylamino)methyl]-2-(4- methoxybenzyl)pyrrolidin-3-ols (1a-m) is described. The pharmacological evaluation of these compounds as anticonvulsants and as neuroleptics was performed. Compounds 1c,d,k and m (30 mg/kg) exhibited 100% protection in mice compared with diphenylhydantoin sodium (DPHS) (50 mg/kg) as a reference drug using maximum pentylenetetrazole seizures test. Compounds 1a,e,f,j and m displayed neuroleptic activity by measuring the latency, ambulation, grooming and rearing parameters using the open field test compared with chlorpromazine as reference drug.
