The 61-69 fragment of H-2Kb molecule, Glu-Arg-Glu-Thr-Gln-Lys-Ala-Lys-Gly is predicted to be a major immunodominant determinant by theoretical considerations.This nonapeptide has been synthesized by liquid phase peptide synthesis.The synthetic (61-69) H-2Kb peptide, when cross-linked to a carrier protein, induced antibodies which bind to the native H-2Kb molecule on the cell surface as assessed by radioimmunoassay.Such antibodies can only be raised in allogenic but not in syngenic strains of mice.Implications of this observation are discussed.
