Design and synthesis of 6-alkyoxyl[1,2,4]triazolo[1,5-a]quinazoline derivatives with anticonvulsant activity

A series of novel 6-alkyoxyl[1,2,4]triazolo[1,5-a]quinazoline derivatives were synthesized in this study. The anticonvulsant activity of all the target compounds 4a-4s, characterized by IR, 1H-NMR and MS, were evaluated using Maximal electroshock test. The pharmacological results showed that some of the compounds displayed positive anticonvulsant activity. Among them, 6-(benzyloxy)-[1,2,4]triazolo[1,5-a]quinazoline (4a) was the most active compound with an ED50 value of 78.9 mg/kg and a PI value of 9.0. Springer Science+Business Media, LLC 2011.

Three-component condensation of 3-amino-1,2,4-triazole (or its 5-methyl and 5-methylthio derivatives), dimedone (or cyclohexane-1,3-dione), and dimethylformamide dimethyl acetal afforded 8,8-R, R-8,9-dihydro[1,2,4] triazolo[1,5-a]quinazolin-6(7H)-ones.
