The invention provides a deuterated piperazine amide compound as well as a preparation method and application thereof. The structure of the deuterated piperazine amide compound is represented by a formula I, wherein in the formula I, R1 is selected from methyl or perdeuterated methyl, and R2-R17 are independently selected from hydrogen or deuterium. The preparation method of the deuterated piperazine amide compound comprises the following steps: reacting a compound A, a compound B and a condensing agent to obtain the deuterated piperazine amide compound. The product provided by the invention has high activity and high safety for treating or preventing neurodegenerative diseases, and is simple in preparation process and suitable for industrial production.
