An adjuvant formulation includes a monophosphoryl Lipid A (MPLA) analogue, a Pam3CSK4 analogue, or a muramyldipeptide (MDP) analogue, or combinations thereof. The adjuvant may be formulated in soluble form or in a nanoparticle, such as polylactic glycolic acid nanoparticles. A vaccine formulation comprises the adjuvant formulation and an immunogen. Novel lipid A mimetics derived from pentaerythritol: Synthesis and their potent agonistic activity

A pentaerythritol unit is substituted for glucosamine as its functional mimic at the reducing end of lipid A disaccharide. Two such pentaerythritol derived lipid A mimetics 3 and 4, carrying three identical fatty acyl moieties, have been designed and synt
