The present invention provides a 2-aminoquinazolinone derivative. The present invention is a compound represented by formula (1) [wherein X1 represents CR or N, X2 represents CR2 or N, X3 represents CR3 or N, Y represents an optionally substituted C6-10 aryl or optionally substituted 6 to 10-membered heteroaryl, Z represents an optionally substituted C6-10 aryl, RA represents a hydrogen atom, halogen, cyano, optionally substituted C1-6 alkyl sulfonyl, optionally substituted C1-6 alkyl, or optionally substituted C1-6 alkoxy, and R1, R2, and R3 independently represent a hydrogen atom, halogen, optionally substituted C1-6 alkyl, or optionally substituted C1-6 alkoxy] or a pharmaceutically acceptable salt thereof.

Abstract: Ortho-selective halogenation of arenes via C–H functionalization has been described under mild reaction conditions. In this reaction Cu(II)X2 was used as a halide source. It is a simple, general and efficient method for the synthesis of 2-halo aromatic isothiocyanates. All the reactions are carried out under optimized reaction conditions to provide their respective desired products in good to high yield. Graphical Abstract: We have developed a general, simple and efficient method for the synthesis of 2-halo arylisothiocyanates from isothiocyanates through ortho-selective halogenation under mild reaction conditions. Cu(II)X2 was used as a halide source for this methodology. All the reactions are carried out under optimized reaction conditions to provide their respective desired products in good to high yield. The present invention relates to tricyclic compounds of formula (I) or pharmaceutically acceptable salt thereof as mPGES-1 inhibitors. These compounds are inhibitors of the microsomal prostaglandin E synthase-1 (mPGES-1) enzyme and are therefore useful in the treatment of pain and/or inflammation from a variety of diseases or conditions, such as asthma, osteoarthritis, rheumatoid arthritis, acute or chronic pain and neurodegenerative diseases.
