The invention discloses a preparation method of 5-azaspiro[2.4]heptane and salt thereof. The preparation method comprises the steps: reacting cyclopropyl nitrile with ethylene oxide under a strong alkaline condition to obtain 1-(2-hydroxy-ethyl)-cyclopropane formonitrile; reducing 1-(2-hydroxy-ethyl)-cyclopropane formonitrile to obtain 2-(1-aminomethyl-cyclopropyl)-ethanol; carrying out a ring closing reaction on 2-(1-aminomethyl-cyclopropyl)-ethanol, to obtain protected 5-(R-)azaspiro[2.4]heptane; and carrying out deprotection on the protected 5-(R-)azaspiro[2.4]heptane to obtain the 5-azaspiro[2.4]heptane or the salt thereof. The used raw materials are cheap, easy to obtain and low in toxicity, meanwhile, the synthesis process steps are few, the reaction operation is simple and convenient, the yield is high, and the method is suitable for large-scale production.
