A convenient one-pot protocol for the synthesis of N,N′-disubstituted and N,N,N′-trisubstituted amidines is reported. In the presence of the Ph3P–I2/Et3N system, a variety of secondary amides were smoothly reacted with primary or secondary amines to afford the corresponding amidines in good to excellent yields under mild conditions.

A series of 14 N2-substituted N1,N1-pentamethylenebenzamidines has been synthesized, their pKa values in 99.8percent, 95.6percent and 50percent ethanol measured and correlated with the Hammett ? constants.The influence of composition of binary mixture ethanol-water within the studied range on pKa values, as well as on ρ values obtained is discussed.It was found that the difference of ρ values obtained for these solvents are small, but remarkable.
