1,4,8,11-Tetraazacyclotetradecane, 1,8-bis[[4-(1,4,8,11-tetraazacyclotetradec-1-ylmethyl)phenyl]methyl]is used as an impurity in the production of Plerixafor (P580500), a hematopoietic stem cell (HSC) mobilizer. Plerixafor is a CXCR4 chemokine receptor inhibitor that blocks the binding of its ligand, stromal cell-derived factor-1-α (SDF-1-α), and is approved for use in combination with granulocyte-colony stimulating factor (G-CSF) to mobilize HSCs to the peripheral blood for collection and subsequent autologous transplantation in patients with non-Hodgkin's lymphoma (NHL) and multiple myeloma (MM).

The synthesis of the four single-ring cyclam derivatives together with the related tri-linked ring derivatives and tetrabenzylated derivative was reported. In the synthesis, differential Boc-protected cyclam derivatives were used as key intermediates. Benzylated and tri-linked cyclam products were found to rely heavily on high-resolution mass spectrometry (HRMS) and all compounds yielded satisfactory HRMS data.
