(5R, 8S, 10R)-8-amino-6-methylergolin derivatives as central dopaminergic stimulating agents

A simplified synthesis of (5R, 8S, 10R)-8-Amino-6-methylergoline, 7, is described. In a systematic variation of this key substance, a series of new derivatives of 7 prepared were tested pharmacologically for central dopaminergic activity. Three substances, 30, 31 and 36, showed high activities in the two screening tests (Ungerstedt-test and observation of stereotyped behaviour) and are further characterized.
