Four novel chiral imidazolium tagged molecules derived from dipeptide (2S,4R)-Hyp-(S)-Phe-OMe were prepared and evaluated as organocatalysts in the asymmetric aldol reaction using water as solvent. It was found that catalysts incorporating the hexanoyl linker are active down to 5 mol% and afford aldol products with up to 94% yield, up to 98:2 dr and up to 97:3 er. This chiral imidazolium catalyst was reused up to 6 times without any loss in the stereoselectivity of asymmetric aldol reactions. By contrast, organocatalysts containing an acetyl linker proved to be highly unstable in protic solvents, and for that reason they could not be recovered and reused.

A straightforward synthetic route to chiral metal-organic frameworks is proposed that relies on an acid-base interaction between an acid linker and a chiral primary amino acid derived diamine organocatalyst. High ee values for the aldol condensation of linear ketones and aromatic aldehydes are reported with this heterogeneous catalyst. Three consecutive catalyst reuse experiments demonstrated that the majority of the activity was preserved, as was the enantioselectivity.

Stable L-proline-functionalized zirconium methyl- and/or phenylphosphonates have been prepared as amorphous solids by the precipitation of (4R)-4-[(4′-phosphonobenzyl)oxy]-L-proline and methyl- and/or phenylphosphonic acid with ZrOCl2. The supported L-proline catalysts were tested in the direct asymmetric aldol addition between several ketones and p-substituted benzaldehydes. High yields, diastereoselectivities (anti/syn up to 92:8) and enantiomeric excesses (up to 99 % ee) have been achieved. Moreover, an easy protocol for the efficient recovery of the catalytic system by simple centrifugation has been defined, and the effective reuse of the catalyst has been reported in the representative aldol addition of cyclohexanone with p-nitrobenzaldehyde. This study has shown that these L-proline immobilized catalytic systems can be used at least six times without loss of activity and with reproducible anti/syn and ee values. Copyright

A series of primary amino acids covalently supported onto polystyrene through alkyne-azide cycloaddition reactions has been synthesized and evaluated as catalysts in asymmetric aldol reactions. A polymer-supported threonine behaves as an easily recyclable, highly reactive and stereoselective (up to 99% ee) catalyst in the aldol reaction of both cyclic and acyclic ketone donors with aromatic aldehydes in aqueous environments. While cyclic ketones react with anti diastereoselectivity, syn adducts are predominantly obtained with acyclic substrates. The heterogenized threonine catalyst has been used for the sequential synthesis of a small library of enantiopure aldol products.
