An unprecedented [2+2+1] hydrative annulation of 1,6-enynes with terminal alkynes is achieved using catalytic cationic Rh(I). Thus, a modular assembly of cyclopenta[b]benzofuranols with two consecutive quarternary stereocenters is achieved from readily available alkynes. The reaction is proposed to go through a sequence of 5-membered rhoda-cycle formation, regioselective acetylene insertion, 1,5 H-shift, substrate controlled stereoselective addition of water molecule followed by 1,2-rhodium migration gave contracted rhoda-cycle D and reductive elimination. Necessary control/labelling experiments were conducted to gain insight in to the mechanism.

Rhodium-catalyzed [2 + 2 + 2] cycloadditions, sulfonyl phthalide annulations and nitroalkene reactions have been employed for the synthesis of 56 quinone-based compounds. These were evaluated against Trypanosoma cruzi, the parasite that causes Chagas disease. The reactions described here are part of a program that aims to utilize modern, versatile and efficient synthetic methods for the one or two step preparation of trypanocidal compounds. We have identified 9 compounds with potent activity against the parasite; 3 of these were 30-fold more potent than benznidazole (Bz), a drug used for the treatment of Chagas disease. This article provides a comprehensive outline of reactions involving over 120 compounds aimed at the discovery of new quinone-based frameworks with activity against T. cruzi.

2,3-Disubstituted indoles bearing 2-hydroxyphenyl moieties at their C3 positions were synthesized from readily available 2-chlorophenols and alkynylanilines via aminopalladation/reductive elimination using Pd–dihydroxyterphenylphosphine catalyst. The catalyst accelerates the introduction of the 2-hydroxyphenyl group at the C3 position of the indole.

The activity of a simple, commercially available copper salt, [Cu(NCMe)4](BF4) in intramolecular hydroamination reactions of alkynes and allenes is presented. Reactions were successfully carried out in technical acetonitrile in the presence of air. While attempts of alkene hydroamination failed, this catalyst was also found active in intermolecular aza-Michael reactions.
