A Modular Construction of Epidithiodiketopiperazines

Epidithiodiketopiperazines (ETPs) possess remarkably diverse biological activities and have attracted significant synthetic attention. The preparation of analogues is actively pursued; however, they are structurally challenging, and more direct and modular methods for their synthesis are desirable. To this end, the utility of a bifunctional triketopiperazine building block for the straightforward synthesis of ETPs is reported. A modular strategy consisting of enolate alkylation followed by site-selective nucleophile addition enables the concise synthesis of (±)-hyalodendrin and a range of analogues.

Structurally diverse natural products from unrelated sources typically require the development of individual synthetic routes. In a novel approach, we have shown that the epidithiodiketopiperazine-derived natural product (±)-hyalodendrin and the core structure of the unrelated pyrrolidine-derived natural product clausenamide can be synthesised from a common synthetic precursor in good yield by simple variation of the reaction conditions. Structurally unrelated natural products often require individual synthetic routes. In our approach, the epidithiodiketopiperazine natural product (±)-hyalodendrin and the core structure of the unrelated pyrrolidine-derived natural product clausenamide were synthesised from a common precursor by simple variation of the reaction conditions.

Disclosed are epidithiodiketopiperazine compounds, pharmaceutical compositions based thereon and methods of treating, reducing or inhibiting transcription and translation of hypoxia-inducible genes. An embodiment of the present invention is directed to a method for interfering with hypoxia-induced transcriptional pathway. Generally, the method according to this embodiment comprises contacting a cell with at least one compound according to either Formula I, Formula II or Formula III, or a salt, solvent or hydrate thereof.

Two general methods, Method A and Method B in Scheme 19, to synthesize epidithiapiperazinediones, are described.A total synthesis of racemic and optically active gliotoxin (1) and of racemic dehydrogliotoxin (53) was achieved by using Method A, whereas a total synthesis of racemic hyalodendrin (52) was completed by using Method B.
