Preparation of labeled bis-amino acid hydroxyethylamino sulfonamide HIV protease inhibitors

Preparation of radiolabeled sulfonamide HIV protease inhibitors, DMP852 and DMP853 is described. The C-14 labeling of DMP852 was accomplished by acylation of the intermediate amine with [1-14C]-chloroacetic anhydride followed by the treatment of the chloroacetyl derivative with methylamine. Due to the rapid loss of the methylglycine moiety during metabolism studies, a route that involved an oxidation-reduction sequence was employed to prepare tritium labeled DMP852.
