Treatment of 2 hydrazino 5 nitrothiazole 1 with methyl isocyanate gave 4 methyl 2 (5 nitrothiazol 2 yl)semicarbazide 4, isomeric with 4 methyl 1 (5 nitrothiazol 2 yl)semicarbazide 3 obtained by the nitration of 4 methyl 1 thiazol 2 ylsemicarbazide 6. Chemical and spectroscopic evidence is given to support the assignments of structure. Congeners of compound 4 were prepared for antibacterial and antiprotozoal screening. Compound 4 and its ethyl homologue were the most active members of the series against Trichomonas vaginalis and Entamoeba histolytica in rodents, and the former possessed high in vitro activity against Gram negative bacteria.
