A series of tridentate ligands for 99mTc, N-(2-hydroxybenzyl)sarcosines (hbs) (2), were prepared by the Mannich reaction of a phenol with formaldehyde and sarcosine.The parent member of the series (2a), for which this method was unsuccessful, was synthesized by reaction of 2-acetoxybenzyl bromide with methyl sarcosinate followed by acid hydrolysis and was isolated as the hydrochloride.
