The dauc-8-en-11-ol synthase from Streptomyces venezuelae was investigated for its catalytic activity towards alternative terpene precursors, specifically designed to enable new cyclisation pathways. Exchange of aromatic amino acid residues at the enzyme surface by site-directed mutagenesis led to a 4-fold increase of the yield in preparative scale incubations, which likely results from an increased enzyme stability instead of improved enzyme kinetics.

Synthesis of 3,7-dimethyl-1-hydroxy-2-E), 6(E)-dodecadien-11-one (1), a component of Mediterranean algae, has been achieved by making use of selenium(IV) oxide/tert-butyl hydroperoxide oxidation and zinc-copper induced conjugate addition using ultrasonic waves. Synthesis of 2,6-dimethyl-2(E),6(E)-octadiene-1,8-diol diisovalerate (2), a sex pheromone of Agriotes tauricus has also been described.

The synthetic utility of the title sigmatropic sequence is illustrated in the syntheses of (E)-6-nonen-2-one, a precursor of exo-brevicomin, and oxocrinol.The scope and limitation of this sequence as a synthetic method for δ,ε-unsaturated ketones are also discussed.
