Pergolide mesylate salt is a potent, long-acting dopamine agonist that is primarily used in the treatment of Parkinson's disease and hyperprolactinemia. It is a dopamine D1 and D2 receptor agonist with high affinity for rat striatal receptors (Kis = 111 and 0.495 nM, respectively). Compared to lergotrile, pergolide mesylate exhibits less toxicity and a lower propensity for inducing psychosis.
Used in Pharmaceutical Industry:
Pergolide mesylate salt is used as an antiparkinsonian agent for its ability to depress dopaminergic firing in paralyzed rats, which can be reversed by dopamine D2-selective antagonist spiperone or dopamine D1-selective antagonist SCH 23390. It is also used to increase the volume threshold for inducing bladder contraction in a cynomolgus monkey model of Parkinson's disease induced by MPTP.
Additionally, Pergolide mesylate salt is used as a dopaminergic agonist to decrease plasma prolactin concentrations, making it useful in the treatment of hyperprolactinemia. It has been shown to reduce carrageenan-induced paw edema in adrenalectomized rats, further demonstrating its therapeutic potential in various conditions.
