The feasibility of the fluoro-olefin function as a peptidomimetic group in inhibitors for dipeptidyl peptidase IV and II (DPP IV and DPP II) is investigated by evaluation of N-substituted Gly-Ψ[CF=C]pyrrolidines, Gly-Ψ[CF=C]piperidines, and Gly-Ψ[CF=C](2-cyano)pyrrolidines. Of this later class, the (Z)- and (E)-fluoro-olefin analogues were prepared and chemical stability in comparison with the parent amide was checked. Most of these compounds exhibited a strong binding preference toward DPP II with IC 50 values in the low micromolar range, while only low DPP IV inhibitory potential is seen.

A method of preparation of α-fluoro α,β-ethylenic carbonyl compounds is described.This method is based on the addition of chlorofluorocarbene, generated at -70 deg C by the action of potassium tertiobutylate on dichlorofluoromethane, to enoxysilanes and a subsequent rearragement of the adducts.With the ketene acetals, α-fluoro α,β-ethylenic esters (or acids) were obtained in high yield by heating the intermediate cyclopropanes in the mixture methanol-triethylamine or in toluene.In the case of trimethylsilyl enol ethers, α-fluoro α,β-ethylenic aldehydes or ketones were obtained in excellent yield after hydrolysis of the siloxycyclopropanes into cyclopropanols and heating of these latter in a non polar solvent in the presence of a tertiary amine.
