Chloroquine is an antimalarial drug which inhibits the DNA and RNA polymerase reactions and which has been shown to bind reversibly with nucleic acids.The "intercalation" mode of binding has been proposed by which the 4-amino-7-chloroquinoline ring of the drug interacts with the nucleotide bases of the polymer.As an approach to the problem of the nature of the forces responsible for these ring-ring stacking interactions, we have prepared models in which the aromatic nucleus of the drug is linked to the nucleotide bases by a trimethylene chain.We describe here the preparation of different models in which the nucleotide bases are adenine and guanine linked through their N-7 atom, and the 4-aminoquinolines bear H or Br at the C-7 position.Comparable compounds in which quinoline is methylated on N-1 are also described.
