The syntheses of 5-deaza-5-oxo analogues of aminopterin, methotrexate, and 10-thiaaminopterin are described.The sequence involves, as a key step, the preparation of 2,4-diamino-6-(hydroxymethyl)-5-oxopyridopyrimidine from readily accessible pyridopyrimidine derivatives.
