Synthesis of 5-Allyloxy-7-hydroxycoumarin

5-Allyloxy-7-hydroxycoumarin (I) has been synthesised by an unambiguous method starting from 6-allyloxy-α-benzyloxy-2-hydroxyacetophenone (IIa), which on cyclisation with diethyl carbonate in the presence of sodium methoxide gives 5-allyloxy-7-benzyloxy-4-hydroxycoumarin (IIIa) along with ethyl 5-allyloxy-7-benzyloxy-4-hydroxycoumarin-3-carboxylate (IVa).Tosylation of IIIa followed by reductive detosylation with Zn-HCl affords I, identical with the natural sample.The formation of coumarin-3-carboxylates has also been observed in the case of 6-benzyloxy-2-hydroxy-4-methoxyacetophenone (IIc) and 4-benzyloxy-2-hydroxy-6-methoxyacetophenone (IId).
