Synthesis and Hypotensive Activity of Some Aminoguanidines

The synthesis of eight heterocyclylaminoguanidines (9b-16b) is described.The compounds possess hypotensive, antihypertensive and adrenolytic activities.Compounds 1,2,3,4-tetrahydroisoquinolin-2-ylguanidine (9b), 1,3-dihydroisoindolin-2-ylguanidine (11b) and 1-(2,3-dihydro-1H-benzisoquinolin-2-yl)guanidine (14b) have been studied in detail.The appearance of tolerance and an unacceptable degree of toxicity prevents further development of the compounds for clinical studies.
