3-Methoxy-8,10-dimethyl-9,10-dihydrophenanthridin-7(8H)-one (III), readily available from 4,6-dimethylcyclohexane-1,3-dione (I), m-anisidine and ethyl orthoformate is condensed with ethyl acrylate to give the adduct (V).The derived acid azide (VII) is submitted to a modified Curtius rearrangement to furnish (+/-)-3-methoxy-11α-6,15-diazaestra-1,3,5,7,9,14-hexaene (VIII), a steroid containing in its framework an 11-methyl group, a quinoline ring and an indole moiety.In another experiment the isoxazole (X) derived from 3-methoxy-10α-methyl-9,10-dihydrophenanthridin-7(8H)-one (IX) is alkylated with methyl iodide and potassium t-butoxide and three products (XI), (XII) and (XIII) methylated to different degree and at different positions have been isolated.The relative configuration and conformation of the hydrophenanthridinones are established from PMR.
