PNGase F, also known as Peptide N-glycosidase F, is an enzyme that cleaves asparagine-linked glycoproteins, resulting in carbohydrate-free peptides and detached full-length oligosaccharides. It has been purified by affinity chromatography and dialyzed into a 50% glycerol solution with 10 mM potassium phosphate pH 7.5 to produce a stable product. The enzyme can be removed from preparative operations by utilizing its C-terminal 6x histidine fusion tag.

83534-39-8 Usage

Uses

Used in Biochemistry Research:
PNGase F is used as a deglycosylation agent for the analysis of glycoproteins, particularly in the study of protein structure, function, and interactions. It helps in the removal of N-linked oligosaccharide chains, allowing researchers to better understand the underlying protein sequences and their roles in various biological processes.
Used in Deglycosylation Assays:
PNGase F from Elizabethkingia meningoseptica is used in deglycosylation assays for various applications, such as evaluating the glycosylation of the Zika virus E protein, verifying the N-linked glycosylation of MHC class 1 polypeptide-related sequence A (MICA), and assessing coxsackievirus and adenovirus receptor glycosylation using CAR-expressing COS cells.
Used in Human Plasma Sample Analysis:
PNGase F has been used in deglycosylation assays in human plasma samples, providing valuable insights into the glycosylation patterns of proteins present in human blood and their potential implications in various diseases and conditions.
Used in Gel, Solution, or Blot Membrane Applications:
The highly purified PNGase F can be used for preparative deglycosylation or for analytical applications in gel, in solution, or on blot membranes, making it a versatile tool for researchers working with glycoproteins in various experimental setups.
Used in Glycopeptidase A-Assisted Deglycosylation:
Glycopeptidase A from almonds, which is used for deglycosylation, works in conjunction with PNGase F to catalyze the removal of N-linked oligosaccharide chains and convert Asn residues to Asp, further enhancing the deglycosylation process and providing a comprehensive approach to studying glycoproteins.

Biochem/physiol Actions

Cleaves an entire glycan from a glycoprotein provided the glycosylated asparagine moiety is substituted on its amino and carboxyl terminus with a polypeptide chain.

Check Digit Verification of cas no

The CAS Registry Mumber 83534-39-8 includes 8 digits separated into 3 groups by hyphens. The first part of the number, starting from the left, has 5 digits, 8,3,5,3 and 4 respectively; the second part has 2 digits, 3 and 9 respectively.
Calculate Digit Verification of CAS Registry Number 83534-39:
(7*8)+(6*3)+(5*5)+(4*3)+(3*4)+(2*3)+(1*9)=138
138 % 10 = 8
So 83534-39-8 is a valid CAS Registry Number.
