Via the condensation of ethyl-2-ethylacetoacetate with substituted 2-aminopyridines in polyphosphoric acid or phosphoryl chloride/polyphosphoric acid, numerous 7-, 8- or 9-carboxylic acid derivatives of 2-methyl-3-ethyl-4-oxo-4H-pyridopyrimidine (6) were synthesized for pharmacological purposes.Several new derivatives of 6 were also prepared by transformations of the functional groups of the pyridine ring.
