Photocyclization of anticoagulant 6-alkyl-4-hydroxy-2-pyrones

Structure-activity relationships of five 4-hydroxy-2-pyrones were investigated by alteration of the alkyl chain at position 6. After oral administration of 330 mg/kg anticoagulant effects were observed only in case of the 6-butyl compound. The 6-ethyl- and 6-butyl compounds underwent photocyclisation to yield cyclobutanes of the α-truxillic acid type, both of which showed no anticoagulant activity.
