Uridine, 5'-deoxy-5-fluoro-5,6-dihydro-, also known as 5-fluoro-5,6-dihydro-5'-deoxyuridine or FUDR, is a fluorinated pyrimidine nucleoside analog. It is a modified version of the naturally occurring nucleoside uridine, with a fluorine atom replacing the hydrogen atom at the 5-position and a reduced double bond between the 5 and 6 positions. FUDR is primarily used as an antineoplastic agent in cancer chemotherapy, particularly for the treatment of liver metastases from colorectal cancer. It works by inhibiting the enzyme thymidylate synthase, which is essential for DNA synthesis, thereby blocking the proliferation of cancer cells. Due to its specific action on rapidly dividing cells, FUDR can also cause side effects in healthy, rapidly dividing cells, such as those in the bone marrow and gastrointestinal tract.
