Synthesis of 3H-[1]Benzofuro[2,3-b]imidazo[4,5-f]quinolines as New Fluorescent Heterocyclic Systems for Dye-Sensitized Solar Cells

Abstract: One-pot reaction of 1-alkyl-5-nitro-1H-benzimidazoles with 2-(1-benzofuran-3-yl)acetonitrile in methanol in the presence of potassium hydroxide gave 3H-[1]benzofuro[2,3-b]imidazo[4,5-f]quinolines as high-performance organic photosensitizers belonging to a new fluorescent heterocyclic system. The structure of the new compounds was assigned on the basis of their spectral and analytical data. Study of the optical properties of the title compounds revealed their interesting photophysical properties such as high fluorescence quantum yields. Their electrochemical properties were studied by cyclic voltammetry, and reversible oxidation waves were observed. The photovoltaic performance of the fluorescent heterocyclic compounds was estimated at 4.89–5.05% from the IPCE spectra and I–V curves.
Synthesis, Antiviral, Antibacterial, and Cytotoxicity Assessment of Some 3H-Benzo[a]imidazo[4,5-j]acridines and 3H-Benzo[a]pyrazolo[3,4-j]acridines

Abstract: Some novel 3H-benzo[a]imidazo[4,5-j]acridines and 3H-benzo[a]pyrazolo[3,4-j]acridines were synthesized by the reaction of 1-alkyl-5-nitro-1H-benzoimidazoles and 1-alkyl-5-nitro-1H-indazoles with 1-naphthylacetonitrile in high yields. The structures of the new compounds were determined by spectral (FTIR, 1H, and 13C NMR) and analytical data. The antiviral activity of the synthesized compounds was tested against a panel of DNA and RNA viruses, including herpes simplex virus-1 KOS, vesicular stomatitis virus, herpes simplex virus-2 (G), vaccinia virus, and herpes simplex virus-1 TK-KOS ACVr. Most of the test compounds showed moderate activities in comparison with their corresponding reference standards. The synthesized compounds were also tested for antibacterial activity against a panel of strains of gram-negative and gram-positive bacterial species, and some of them we found as effective against gram- positive bacteria as well-known antibacterial agents, such as Cephalexin. The products we found to be cytostatic in the higher micromolar range.
Interactions of human serum albumin with bioactive 3H-imidazo[4,5-a]acridines: Insights from fluorescence spectroscopic studies

Several 3H-imidazo[4,5-a]acridine derivatives were conveniently synthesized by the reaction of imidazo[4,5-a]acridones in boiling POCl3. The imidazoacridones were obtained by rearrangement of 3H-imidazo[4′,5′:3,4]benzo[c]isoxazoles in concentrated sulfuric acid containing nitrous acid at room temperature. The structures of all newly synthesized compounds were confirmed by IR, 1H NMR, and mass spectral data. The interactions of 3H-imidazo[4,5-a]acridines with human serum albumin (HSA) were studied by fluorescence spectroscopy. The binding of 3H-imidazo[4,5-a]acridines quenches the HSA fluorescence, revealing a 1: 1 interaction with a binding constant of about 2.34 × 105-3.16 × 106 M-1. A decrease in fluorescence intensity at 339 nm, when excited at 280 nm, is attributed to changes in the environment of the protein fluorophores caused by the presence of the ligand. The differences in interactions of 3H-imidazo[4,5-a]acridines with HSA were observed using spectrofluorimetry technique.

Resistance to antimicrobial agents is a concern that exists globally and has a considerable impact on human and animal health, so that the discovery of new antibacterial compounds has become increasingly more important in combating infectious disease. In this paper, imidazo[4,5-a]quinindolines are introduced as new antibacterial agents against Gram-positive and Gram-negative bacteria. These pentacyclic compounds are synthesized by the reaction of N-alkyl-5-nitrobenzimidazoles with 2-(1-alkyl-1H-3-indolyl)acetonitrile under basic conditions in excellent yields. The structures of newly synthesized compounds were characterized by IR, 1H NMR, 13C NMR, and mass spectral data. The antibacterial activities of the synthesized compounds were screened against standard strains of two Gram-positive and two Gram-negative bacteria using the broth microdilution method. Most of the compounds studied showed promising activities against both types of bacteria.
Synthesis, Spectroscopic Characterization and DFT/TD-DFT Calculations of new Fluorescent Derivatives of Imidazo[4′,5′:3,4]Benzo[c]Isoxazole

An increasingly wide variety of fluorescent compounds is used in biotechnology, genomics, immunoassays, array technologies, imaging, and drug discovery. Therefore, synthesis of fluorophores with novel structural features can be interesting and useful in various fields. In this paper, four new fluorescent heterocyclic compounds with high quantum yields are introduced. These new fluorophores are synthesized in moderate to high yields via regioselective nitration of 3-alkyl-8-(4-chlorophenyl)-3 H-imidazo[4′,5′:3,4]benzo[c]isoxazoles. The latter compounds are obtained from the reaction of 1-alkyl-5-nitro-1 H-benzoimidazoles with (4-chlorophenyl)acetonitrile in basic MeOH solution. Physical spectral (UV-vis, IR, 1HNMR, 13C NMR, NOESY and fluorescence) and analytical data have established the structures of synthesized compounds. The fluorescence properties of new fluorescent heterocyclic compounds are studied. The fluorescence of all compounds is very intense and fluorescence quantum yields are high (> 0.52). Density functional theory (DFT) calculations are performed to provide the optimized geometries, relevant frontier orbitals and the prediction of 1H NMR chemical shifts for confirming the exact structure of fluorescent compounds. Calculated electronic absorption spectra were also obtained by time-dependent density functional theory (TD-DFT) method.
