Bovine serum albumin (BSA) is a water-soluble protein derived from the blood of cows, playing a key role in regulating blood colloidal osmotic pressure. It is composed of 583 amino acids with a calculated molecular weight of 66,430 Daltons. BSA is the major protein in bovine serum and has six α-helices forming three homologous domains. Depending on pH, BSA undergoes reversible conformational isomerization, making its native structure reactive and flexible upon heating.


Used in Molecular Biology Applications:
Bovine albumin is used as an enzyme or protein stabilizer for preventing adhesion to reaction tube surfaces or pipette surfaces, and as an inert stabilizer during incubation to allow proper enzyme or protein function.
Used in Immunoassays:
Bovine albumin is used as a blocking agent to minimize non-specific antibody binding in immuno-detection procedures such as western blotting, immunofluorescence, and immunohistochemistry (IHC).
Used in Cell Culture Media:
Bovine albumin is used as a supplement in cell culture media to provide essential nutrients for cell growth and maintenance.
Used in Protease-Sensitive Assays:
Bovine albumin is used as a stabilizer for enzymes during digestion of DNA and to prevent adhesion of the enzyme to reaction tubes and other vessels. It is also used in protease-sensitive immunoassays such as RIA, EIA, fluorescent and chemiluminescent assays, and as a protein standard, diluent, or conjugate.
Used in Diagnostic Applications:
Bovine albumin is used as a constituent of medium for the differentiation of human pluripotent stem cells, in enzyme-linked immunosorbent assay (ELISA), and sandwich ELISA. It has been specifically used as a component of blocking buffer in a peptide-based ELISA assay for analysis of COVID-19 / SARS-CoV-2 samples.
Used in Research Applications:
Bovine albumin is used in various research applications, including protein quantitation as a standard, immunochemistry as a blocking agent, and cell culture as a nutrient. It is also used in enzyme assays, protein-based assays, and protease-sensitive techniques such as enzyme immunoassay (EIA), nucleic acid hybridization, and radioimmunoassay (RIA).
Used in Positive Controls:
Acetylated BSA may be used as a positive control for ELISA or Western Blot when employing an acetyl-lysine monoclonal or polyclonal antibody, particularly for detecting histone subunit modifications such as lysine acetylation.
