The double-oxidative dehydrogenative (DOD) cyclization represents one of the most straightforward and atom-economical methods for cyclic structure formation. A Cu(II)/DDQ/O2 system-catalyzed DOD [4 + 2]-annulation/oxidative aromatization tandem reaction of readily available glycine derivatives and alkylbenzenes was established. This approach facilitates rapid access to a broad scope of substituted quinoline-2-carboxylate derivatives, an important motif in drug discovery. The reaction could feasibly be applied to a 10 gram-scale synthesis.

The invention provides a method for synthesizing quinoline compounds through a double-oxidation dehydrocyclization reaction. The method comprises the following steps: with N-arylglycine derivatives, ethylbenzene and derivatives thereof as raw materials, copper salt as a catalyst and DDQ as a dehydrogenating agent, reacting at 60-90 DEG C for 5-18 hours under the action of an oxidant; after completion of reaction, performing column chromatography separation to obtain target products. By the method, reaction reagents are low in price and easy to obtain, reaction steps are simple and short, a synthesis condition is mild, the reaction speed is high, the reaction yield is relatively high, the product purity is good, posttreatment is simple and convenient and the industrial production cost can be effectively reduced; therefore, the method is suitable for industrial production.
