The synthesis of ethyl 3,5-dimethyl-4-isothiazolylacetate, 1, and the corresponding nitrile 2 as well as (3,5-dimethyl-4-isothiazolyl)methyl p-tolyl sulphone, 3 , are reported.These compounds react with different bases yielding carbanions at C4α, which are transformed into 4α-substituted isothiazoles by reaction with electrophiles in alkylation, acylation, 1,2-addition or Michael-type processes.The behaviour of the intermediates in these reactions is strongly dependent on the steric requirements at C4α.
