4-Demethoxydaunorubicin (3), a new synthetic antitumor anthracycline under clinical evaluation, was stereoselectively converted to the corresponding 13-(R)-dihydro derivative (4-R) by using a blocked mutant of Streptomyces peucetius.The absolute configuration of the C(13) of the new anthracycline 4-R was assigned on the basis of 1H-NMR examination of the 9,13-O-isopropylidene derivative of its aglycone 5-R in comparison with the corresponding C(13) epimers obtained by chemical reduction.Compound 4-R was also identified as the major human urinary metabolite of 3.
