Condensation of 1,2,3,4,6,7,8,9-octahydro-9aH-quinolizin-2-one (4) with triethylphosphonoacetate gives a mixture of 2-ethoxycarbonylmethylene-1,2,3,4,6,7,8,9-octahydro-9aH-quinolizin-2-one (5a) and ethyl 1,4,6,7,8,9-hexahydro-9aH-quinolizine-2-acetate (5b) which on catalytic reduction affords the required ester 6.Condensation of 6 with 3,4-dimethoxyphenethylamine followed by cyclisation with POCl3 and reduction with NaBH4 furnishes the title compound 10.The amide 7 on LiAlH4 reduction gives the 1',8'a-seco analog (9) of 10.These compounds have been evaluated for their antiamoebic, antileishmanial and anticancer activities.
