DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) In freely moving cats, LSD [50-37-3] (10, 50, and 100 mg/kg, i.p.) produced a dose-dependent decrease in the nucleus centralis superior (NCS) unit activity. The drug also produced a dose-dependent increase in certain behaviors (e.g., limb flicking and abortive grooming) and the peak behavioral and unit changes were temporally correlated. By contrast, LSD had little effect on serotonin-contg. nucleus raphe pallidus (RPA) neurons. 5-Methoxy-N,N-dimethyltryptamine (5-MeODMT) [1019-45-0] also produced a dose-dependent decrease in NCS unit activity at 10, 50 and 250 mg/kg (i.m.) and dose-dependent behavioral changes. 5-MeODMT had no overall significant effect on RPA unit activity, except at the highest dose level. Psilocin [520-53-6] produced dose-dependent decreases in NCS unit activity at 25, 100 and 750 mg/kg (i.p.), whereas the behavioral changes were not dose-related. Psilocin also had relatively little effect on the activity of RPA neurons. The phenylethylamine hallucinogens, 2,5-dimethoxy-4-methylamphetamine [15588-95-1] (50, 250 and 1000 mg/kg i.p.) and mescaline [54-04-6] (5000 mg/kg i.p.), both produced large behavioral changes, but no overall significant effect on raphe unit activity in either the NCS or RPA. Ascending dorsal raphe and NCS neurons may be involved in the process of hallucinogenesis, whereas descending RPA neurons do not appear to be involved in this process. The psychol. and perceptual effects of hallucinogens are attributable to an inactivation of central serotonergic neurotransmission.

DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Serotonergic neurons were recorded in the nucleus centralis superior (NCS) in freely moving cats and were initially identified on-line by their slow and regular spontaneous activity (mean 2.55 spikes/s). Discharge rates of NCS serotonergic neurons were highest during active waking (AW) (mean 2.94 spikes/s), decreased during slow-wave sleep (middle of SWS: mean 1.38 spikes/s), and were lowest during rapid-eye-movement sleep (mean 0.46 spikes/s). The activity of serotonergic NCS neurons did not increase during transient elevations of the electromyog. activity during AW but did decrease immediately preceding, and during the occurrence of, SWS spindles. These neurons were responsive to phasic auditory and visual stimuli, with most neurons showing excitatory responses. In response to a small dose of the serotonin [50-67-9]-specific agonist 5-methoxy-N,N-dimethyltryptamine [1019-45-0] (50 mg/kg, i.m.), NCS serotonergic neurons responded with a mean decrease in unit activity of 43.9%. Among the NCS serotonergic neurons a subpopulation differed from the remaining serotonergic neurons in that they showed a much smaller decrease in unit activity across the sleep-wake cycle and responsed with an inhibition of activity to phasic auditory and visual stimuli. The results are compared with those previously reported for serotonergic neurons in the dorsal raphe nucleus, nucleus rape pallidus, and nucleus raphe magnus of freely moving cats.
