DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) After administration of 10 or 25 mg d,l-methadone-HCl (I) [1095-90-5] to adult female albino guinea pigs, the b-phase plasma half-lives were 13.2 and 13.8 h, resp. Animals treated with 6 consecutive daily doses of I showed peak plasma and hepatic levels by day 2 of the treatment followed by a gradual decline on subsequent days. I did not initially affect either hepatic monooxygenase [9038-14-6] or aniline hydroxylase [9012-80-0] activities. Significant decrease of glucuronyltransferase [37329-64-9] activity was obsd. 24 h after the initial dose. No overt toxicity was obsd. with I administration.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Body wt. gain and food and water consumption of female rats receiving daily methadone-HCl [1095-90-5] injections (5 mg/kg) i.p. prior to mating and during gestation and lactation were studied. In contrast to saline-injected controls, methadone-treated females had lower body wts. throughout pregnancy, but gained wt. during lactation; by postnatal day 21, both groups were of comparable body wt. No significant differences in food consumption throughout gestation or lactation were recorded. Total water consumption of methadone-treated rats was higher during the first 2 wk of gestation and lactation, and lower during the 3rd week of lactation when compared to controls. Thus, methadone-treated rats have lower body wts. throughout gestation and lactation, and that these wt. deficits are not caused by insufficient food and water consumption.
