DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Expts. were carried out to study the effects of 4 bile acids, chenodeoxycholic acid (CDCA) [474-25-9], deoxycholic acid (DCA) [83-44-3], ursodeoxycholic acid (UDCA) [128-13-2], and cholic acid (CA) [81-25-4], on the leakage of glutamic oxalacetic transaminase (GOT) and lactic dehydrogenase (LDH) from isolated rat hepatocytes, and on hypotonic hemolysis using rat erythrocytes. The surface activity of these bile acids was also measured. The same study was also conducted on alkyltrimethylammonium salts-decyltrimethylammonium bromide (C10) [2082-84-0], dodecyltrimethylammonium bromide (C12) [1119-94-4], tetradecyltrimethylammonium bromide (C14) [1119-97-7], and hexadecyltrimethylammonium bromide (C16) [57-09-0]. A close correlation between their membrane effects and their surface activities was found. These membrane effects varied with the no. and the place of the hydroxyl group in the hydrophilic region of bile acids, and the length of their alkyl chains in the hydrophobic region in alkyltrimethyl ammonium salts. The order of the strength of the bile acids was CDCA3 DCA > UDCA3CA, and in the alkyltrimethylammonium salts was C163C14> C12>C10. The surface activities of these compds. seem to be related to their ability to induce hepatic damage.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The actions of 6 members of the homologous series of N-alkyltrimethylammonium ions CH3(CH2)n-1N+(CH3)3(CnTMA) on the Na current in giant axons of Loligo forbesi were investigated. The substances tested correspond to n = 6, 8, 10, 12, 14 and 16. These cations only produced significant Na current suppression when applied inside the axon. Actions on first-pulse Na currents and use-dependent effects were sep. studied. The shorter members of the series (C6TMA [2650-53-5] and C8TMA [2083-68-3]) produced suppression of first-pulse Na currents without causing significant use dependence. The first-pulse suppression arose partly from a pos. shift along the voltage axis of the steady-state activation parameter (m￥) and partly from a redn. in the max. Na conductance. C12TMA [1119-94-4] and C14TMA [1119-97-7] produced little first-pulse suppression but caused clear use dependence. C10TMA [2082-84-0] showed intermediate properties while C16TMA [57-09-0] was inactive. The use-dependent actions have been quant. investigated using a double-pulse protocol. The results are consistent with a model in which the cations enter a blocking site on the ion-channel via the intra-axonal aq. phase. The cations appear able to bind to inactivated Na channels at significant rates. The possible mol. locations of the sites responsible for m￥ shifts and use dependence are discussed. It is argued that the existence of 2 sep. sites may help to explain certain distinctions between the actions of neutral general anesthetics and clin. local anesthetics on the Na channel.
