- Metabolism of phosphoric acid triesters by rat liver homogenate
- Metabolism of phosphoric acid triesters by rat liver homogenate. Sasaki, Kumiko; Suzuki, Takashi; Takeda, Mitsuharu; Uchiyama, Mitsuru (Natl. Inst. Hyg. Sci., Tokyo, Japan). Bull. Environ. Contam. Toxicol., 33(3), 281-8 (English) 1984. CODEN: BECTA6. ISSN: 0007-4861. DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Rat liver microsomes possessed enzymes decompg. tri-Bu phosphate (TBP) [126-73-8], tris(1,3-dichloroisopropyl) phosphate (TDCPP) [13674-87-8], and tri-Ph phosphate (TPP) [115-86-6] in the presence of NADPH [53-57-6]. TBP was most readily decompd., followed by TPP, and then TDCPP. In the absence of NADPH, only TPP was easily decompd. (66%) whereas TBP (11%) and TDCPP (7%) were not. Tris(2-chloroethyl) phosphate [115-96-8] was scarcely metabolized by rat liver. TBP was easily oxidized to TBP-OH [89197-69-3] and further to TBP-(OH)2 [89197-71-7] by microsomal mixed-function oxidase (MFO) [9040-60-2]. TDCPP was decompd. partly by MFO to bis(1,3-dichloroisopropyl) phosphate [72236-72-7], and metabolized more efficiently to the glutathione conjugate [93552-72-8] by the sol. fraction. TPP was decompd. to di-Ph phosphate [838-85-7] by MFO and arylesterase [9032-73-9] in microsomes.
