DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Both metoclopramide (I) [364-62-5] and metopimazine (II) [14008-44-7] had intestinal-spasmolytic activity in mice, as shown by their slowing the intestinal transit of a test meal. At equal doses, II was more effective than I; the latter, when given i.p., was 0.1 as active as atropine. Tests on the isolated guinea pig ileum also showed both I and II to be spasmolytic. I inhibited the contractions induced by acetylcholine, histamine, and BaCl2, indicating both a neurotropic and a musculotropic action. II, on the other hand, was neurotropic only. The mean effective concn. of both drugs was 50 .mu.g/mL for blocking the effects of acetylcholine chloride and histamine-HCl at 10 .mu.g/mL.

I increased the respiratory rate in cats and rabbits after i.v. injection but inhibited respiration and decreased blood pressure when administered intracisternally. The pharmacol. effects of I were, in general, less than those of chlorpromazine.
